Item type:Publication, Morphine self-administration is inhibited by the antioxidant N‐acetylcysteine and the anti-inflammatory ibudilast; an effect enhanced by their co-administration(2024) ;María Elena Quintanilla ;Paola Morales ;Daniela Santapau ;Javiera GallardoRocío Rebolledo<jats:sec id="sec001"> <jats:title>Background</jats:title> <jats:p>The treatment of opioid addiction mainly involves the medical administration of methadone or other opioids, aimed at gradually reducing dependence and, consequently, the need for illicit opioid procurement. Thus, initiating opioid maintenance therapy with a lower level of dependence would be advantageous. There is compelling evidence indicating that opioids induce brain oxidative stress and associated glial activation, resulting in the dysregulation of glutamatergic homeostasis, which perpetuates drug intake. The present study aimed to determine whether inhibiting oxidative stress and/or neuroinflammation reduces morphine self-administration in an animal model of opioid dependence.</jats:p> </jats:sec> <jats:sec id="sec002"> <jats:title>Methods</jats:title> <jats:p>Morphine dependence, assessed as voluntary morphine self-administration, was evaluated in Wistar-derived UChB rats. Following an extended period of morphine self-administration, animals were administered either the antioxidant N-acetylcysteine (NAC; 40 mg/kg/day), the anti-inflammatory ibudilast (7.5 mg/kg/day) or the combination of both agents. Oxidative stress and neuroinflammation were evaluated in the hippocampus, a region involved in drug recall that feeds into the nucleus accumbens, where the levels of the glutamate transporters GLT-1 and xCT were further assessed.</jats:p> </jats:sec> <jats:sec id="sec003"> <jats:title>Results</jats:title> <jats:p>Daily administration of either NAC or ibudilast led to a mild reduction in voluntary morphine intake, while the co-administration of both therapeutic agents resulted in a marked inhibition (-57%) of morphine self-administration. The administration of NAC or ibudilast markedly reduced both the oxidative stress induced by chronic morphine intake and the activation of microglia and astrocytes in the hippocampus. However, only the combined administration of NAC + ibudilast was able to restore the normal levels of the glutamate transporter GLT-1 in the nucleus accumbens.</jats:p> </jats:sec> <jats:sec id="sec004"> <jats:title>Conclusion</jats:title> <jats:p>Separate or joint administration of an antioxidant and anti-inflammatory agent reduced voluntary opioid intake, which could have translational value for the treatment of opioid use disorders, particularly in settings where the continued maintenance of oral opioids is a therapeutic option.</jats:p> </jats:sec>1Scopus© Citations 3 - Some of the metrics are blocked by yourconsent settings

Item type:Publication, New Highly Portable Simulator (SECMA) Based on Virtual Reality for Teaching Essential Skills in Minimally Invasive Surgeries<jats:p>This study presents a new minimal access surgery training system, SECMA, and its constructive validation to determine its usefulness for training basic laparoscopic skills. SECMA is an affordable, highly portable, mobile virtual reality training tool for laparoscopic techniques that integrates the Oculus Quest with a mechanical interface for surgeon simulation of forceps using the hand controllers of these devices. It allows the execution of structured activities (supported by virtual scenarios simulating operating rooms developed in Unity), performance evaluation, and real-time data capture. Two experiments were carried out: 1) coordination; and 2) capture and transport, with a total of 21 individuals divided into two groups: a novice group (inexperienced) of 10 participants and an expert group (>100 endoscopic procedures) of 11 participants. Total task time score, right-hand speed, path length, and other metrics from several consecutive runs on the simulator were compared between experts and novices. Data automatically recorded by SECMA during the experiments were analyzed using hypothesis tests, linear regressions, analysis of variance, principal component analysis, and machine learning-supervised classifiers. In the experiments, the experts scored significantly better than the novices in all the parameters used. The tasks evaluated discriminated between the skills of experienced and novice surgeons, giving the first indication of construct validity for SECMA. </jats:p>11Scopus© Citations 2 - Some of the metrics are blocked by yourconsent settings
Item type:Publication, SimText: a text mining framework for interactive analysis and visualization of similarities among biomedical entities(2021) ;Marie Macnee; ;Sarah Schumacher-Bass ;Jarrod DaltonCostin Leu<jats:title>Abstract</jats:title> <jats:sec> <jats:title>Summary</jats:title> <jats:p>Literature exploration in PubMed on a large number of biomedical entities (e.g. genes, diseases or experiments) can be time-consuming and challenging, especially when assessing associations between entities. Here, we describe SimText, a user-friendly toolset that provides customizable and systematic workflows for the analysis of similarities among a set of entities based on text. SimText can be used for (i) text collection from PubMed and extraction of words with different text mining approaches, and (ii) interactive analysis and visualization of data using unsupervised learning techniques in an interactive app.</jats:p> </jats:sec> <jats:sec> <jats:title>Availability and implementation</jats:title> <jats:p>We developed SimText as an open-source R software and integrated it into Galaxy (https://usegalaxy.eu), an online data analysis platform with supporting self-learning training material available at https://training.galaxyproject.org. Item type:Publication, The Impact of Flashing on the Efficacy of Variable Message Signs: A Vehicle-by-Vehicle Approach(2022) ;Franco Basso ;Pedro Maldonado ;Raúl Pezoa ;Nicolás Szoloch<jats:p>A great deal of research has examined the efficacy of variable message signs (VMS) to induce driver behavior changes, improve safety conditions, and decongest the traffic network. However, there is little literature regarding the most effective ways to display this information on VMS. Furthermore, none of the previous contributions have concentrated on analyzing what impact flashing VMS have on drivers by using real traffic data. This article seeks to bridge this gap, analyzing the effect of incorporating intermittent light stimulation to messages on drivers’ behavior on a Chilean highway, using vehicle-by-vehicle data obtained in a non-intrusive way. In order to do so, an experiment was carried out to measure the responses of drivers when faced with two types of messages: (1) those intended to induce a speed reduction and (2) those aimed at generating lane changes. From the statistical models we obtained several insights. Our results show that flashing messages may increase the effectiveness of VMS depending on environmental and traffic conditions. In particular, for speed moderation messages, we found 12 significant effects, showing, for example, that a flashing message is most effective in the hours of darkness, with low congestion, small spacing, and low average speeds. Additionally, it has a more significant impact on experienced drivers. On the other hand, for lane change messages, we found five significant effects, showing that flashing messaging reduces its effectiveness in situations where a high cognitive load is required, such as in high flow and high average speeds.
